How to grow spores

Inoculating a sterile grain bag with a 10cc spore syringe is a common method for cultivating mushrooms. Here's a step-by-step guide on how to do it:

Materials needed:

  • Sterile grain bag

  • 10cc spore syringe

  • Alcohol swabs or a spray bottle with isopropyl alcohol

  • A clean work area

  • Gloves and face mask

  • A heat source (such as a pressure cooker or a pot of boiling water)

Steps:

  1. Sterilize the grain bag and syringe: Start by sterilizing both the grain bag and the syringe. This can be done by heating them in a pressure cooker or boiling water for about 30 minutes. Make sure the syringe is cooled down before use.

  2. Prepare your work area: Set up a clean work area, and put on your gloves and face mask. Spray or wipe down the work area with alcohol to disinfect it.

  3. Inject the spores: Take the syringe and inject the spore solution into the grain bag. Make sure to inject the spores evenly throughout the bag to ensure even colonization. A good rule of thumb is to inject 1cc of spore solution per 100cc of grain.

  4. Seal the bag: After injecting the spores, seal the bag by folding the top of the bag over a few times and then securing it with a clip or tape. This will keep the contents of the bag sterile while the spores colonize the grain.

  5. Incubate the bag: Place the grain bag in a warm and dark place, such as a closet or incubator, and let it sit for about two weeks. During this time, the spores will germinate and begin to colonize the grain.

  6. Monitor the bag: After two weeks, check the bag for signs of colonization. This will look like white, fuzzy growth spreading throughout the grain. If the bag is not fully colonized, let it sit for another week or so until it is.

  7. Use the colonized grain: Once the grain is fully colonized, it can be used to inoculate other growing mediums, such as substrate bags or trays, to produce mushrooms.
